I would have to go for Chepito from Grim Fandango... I still get that dumb song stuck in my head sometimes... "hide it under some seaweed? NO!! let it shine let it shine etc..."
But I also liked Potts in Steel Sky... what an anal (place expletive here).
He confiscates all your stuff, and I spent a good portion of the game trying to figure out how to get it all back, and then it turns out to be a wild goose chase. Great stuff.

Erana from QFG. :)
Jelena

Broken Sword III:
I wouldn't call her a favourite, but this woman in curlers sitting outside/managing a 'pissoir' is hilarious.

I'm rather fond of Elaine Marley, myself.
Glottis is definitely up there, though.
But I also liked Potts in Steel Sky... what an anal (place expletive here).
Is that the mechanic dude at the start?
He's great, if only for:
"I thought about RE-DECORATING... but I couldn't be ARSED!"
Well, it works better on the talkie version.
I'm quite partial to Sophia on FoA now, but that's mainly because I've just replayed it. The interaction between her and Indy is a lot of fun, and the designers have gone to great lengths to give you something contextually relevant to your current situation to discuss - usually completely optional.
You do get to 'play' her at odd points though, so perhaps she's not a true NPC!

Oscar in Syberia
Boos Miller "Want some Rye?" in (Return To Zork)
Once A Villain

He would also be in a villain thread, but it would have to be Von Glower.

Does this have to be limited to adventures? Because my favorite NPC is from an RPG.

No doubt about it; LeChuck.
Also the narrator from Space Quest 4

Kenny, the balloon-animal guy from LSL7.
Kenny: "I make balloon animals for all the children aboard the ship!"
Larry: "Uh, I haven't seen any children on this ship..."
Kenny: "Of course not, silly! This game is TOOOOO filthy for children!!"
the guy just cracked me up. Plus, the different animations of him making his "animals" fight each other, the way he "held" one balloon animal in order to make another one...goofy as hell, makes me laugh just visualizing all of it.
In a non-humorous game, I'd say Nemesis from Zork: Nemesis. He was just sooooo creepy.

Cedric the owl from King's Quest 5. "Look Graham, there's a boat here...maybe we can uuuuuuuuuuuuuuuuuuuuuse it!"
Kidding. In all seriousness, I think that the Gabriel Knight games have some of the best developed and interesting supporing characters. Grace in GK1 (you can't play as her), Detective Mosley, Gerde, Wolfgang, Emilio and Von Glower are a few of my favorites.
Captain Blondebeard

Largo in MI2 and Murray in MI 3,4

Detective Mosley
Good choice, mine too. :D
My other is Murray the talking demonic skull from MI 3 & 4.

Yeah, Mosley from GK1 stood out to me immediately. (In fact, I'm immediately changing my avatar in honor of him - too many Gabriel's and no love for the real hero of the first game)
Glottis and Cortez from TLJ also were very cool and original characters.
Wonkers from Dreamfall may be the best side character character with so little screentime and purpose in the game. Just kind of a small background character for the very beginning of the game, and yet he makes such an impact. I wish we could play a game with Wonkers before leaving him. :frown:
Can't go past Lemonhead.
God bless the Vegetarian Cannibal!!! loved him whenever he made an appearance, such a typical monkey island funny character... epitomises the whole MI ideal.

No one mentioned Joey from Beneath A Steel Sky, he's one of the best thing in the game (actually, the game itself is not that great imo, unlike some reviews of it I've read. If it wasn't for Joey and the cool accents in the talkie version I would've probably considered it a failure).
Momo from Syberia. I wonder what became of him.
Boneho Chane

Dwane from BS1 is very funny.

When thinking of a favorite NPC, adventures really don't come to mind. But since this is, after all, the Adventure forum, I'll go with Glottis. Not because I have any particularly fond memories of Glottis other than him sitting by the piano singing, "Woah woah woah, woah woah woah, Bone... waGUNNN!!!", but just because I really can't think of any other adventure game NPCs I have even that many fond memories of.
Edit: Okay, to be fair, he did have one great line: "We'll soar like eagles! EAGLES ON POGO STICKS!"

My favourite non-playing character is that whistling Irish kid from Broken Sword - McGuire. Dunno why but he just made me laugh.
Oh yeah and Stan from Monkey Island 1-4 - he's awesome

That kid in front of the pub in Black Mirror was funny, you looked at him and Samuel would say "he likes chocolate" and eventually...
HE DIED! To be honest, that was a good bit of the game because he was brutally sacrificed the annoying git.

The NPCs in Tim Schafer's adventures are all wonderful.
And Mephisto (Mef) in Faust was a superb character.

Von Glower comes to mind, of course..
Otherwise, well, all the characters in Callahan's Crosstime saloon are great, same goes for Monkey Island (Stan is just amazing).
And there's also Kerner. :)

Floyd from Planetfall is my all-time favourite character.

Mosley! Mose, Mose, Mose. Some where, in Sierra-Jensen land, you're still rockin' the gold blazer. Keep on truckin', dude.
